Boulder biotech Enveda announced Phase 1 clinical trial results for ENV-308, an experimental oral drug designed to mimic the weight management and metabolic effects of exercise. Tested on 88 healthy adults, the once-daily pill demonstrated safety, good tolerability, and a favorable gastrointestinal profile, positioning it as a distinct alternative or complement to GLP-1 medications.

Phase 1 Results and Safety Profile of ENV-308
The initial human study enrolled 88 healthy adults who received varying doses of the experimental drug. Rather than testing efficacy or weight loss, researchers focused primarily on safety and tolerability. Participants tolerated the medication across the full dosage range, and the company reported an exceptional gastrointestinal safety profile.
Gastrointestinal distress such as nausea and vomiting frequently leads patients to discontinue blockbuster GLP-1 medications like Ozempic and Wegovy. During the Phase 1 trial for ENV-308, no participants experienced serious adverse events or stopped taking the medication due to side effects. Researchers also tracked circulating levels of leptin, a hormone released by fat cells to control appetite. The trial revealed that the pill reduced circulating leptin levels in human volunteers, mirroring effects observed in preclinical animal models where the drug rapidly lowered elevated leptin associated with obesity.
How ENV-308 Mimics Lac-Phe Chemistry
The underlying science stems from the discovery of Lac-Phe (lactate phenylalanine), a small molecule metabolite identified by Stanford scientists in 2022. The human body naturally produces Lac-Phe when lactate—released by active muscles during vigorous exercise—combines with the amino acid phenylalanine. Once in the bloodstream, Lac-Phe signals the brain to suppress appetite.
While natural Lac-Phe breaks down too rapidly in the body to maintain sustained appetite suppression, Enveda utilized its proprietary AI model PRISM to discover ENV-308 as a stable oral alternative.

Beyond differing pathways, animal studies indicate that ENV-308 preserves lean muscle mass during weight loss and helps maintain reduced body weight after cessation. Muscle preservation addresses a notable concern with traditional weight-loss regimens, where patients frequently experience lean tissue reduction alongside fat loss. Dr. Paul Sack, an endocrinologist at MedStar Health, noted that while animal findings are promising, human data showing weight loss effectiveness is not yet available.
Future Clinical Development and Potential Combinations
Experts suggest the medication may find its primary utility alongside existing therapies rather than as a standalone replacement.
